Repurposing of Acriflavine to Target Chronic Myeloid Leukemia Treatment

Drug repurposing has lately received increasing interest in several diseases especially cancers, due to its advantages facilitating the development of new therapeutic strategies, by adopting a cost-friendly approach and avoiding strict Food Administration (FDA) regulations. Acriflavine (ACF) is an FDA approved molecule that been extensively studied since 1912 with antiseptic, trypanocidal, anti-viral, anti-bacterial anti-cancer effects. ACF shown block growth solid hematopoietic tumor cells. Indeed, acts as inhibitor various proteins, including DNA-dependent protein kinases C (DNA-PKcs), topoisomerase I II, hypoxia-inducible factor 1α (HIF-1α), addition recent discovery signal transducer activator transcription (STAT). Chronic myeloid leukemia (CML) clonal myeloproliferative disorder characterized expression constitutively active tyrosine kinase BCR-ABL. This allows activation signaling pathways known for their role cell proliferation survival, such JAK/STAT pathway. CML therapy, based on inhibitors (TKIs), imatinib (IM), highly effective. However, 15% patients are refractory IM, where some cases, 20-30% become resistant. Thus, we suggest after IM failure or combination improve anti-tumor effects IM. In this review, present different pharmacological properties along anti-leukemic hope therapy.
